New Innovation at Hupo
As we get ready for this year’s HUPO 2022 in Cancun, Mexico, we can already reveal that we will feature a programme filled with new innovations, scientific presentations, and networking opportunities.
We look forward to announcing a new high throughput method as well as automated workflows which are critical to large cohort studies and ultimately bringing proteomics to the clinic.
For a widespread adaptation of proteomics in the clinic, the entire workflow from sample preparation through to data analysis needs to be fast and robust to enable the required throughput. Therefore, we introduce a fully automated end-to-end sample preparation workflow starting from protein to tryptic peptides loaded on Evotips and applied to a clinical cohort of patient plasma samples.

Mass spectrometry-based clinical proteomics is becoming increasingly popular and provides a powerful set of technologies, with the potential to revolutionize health care and enable precision medicine. We specifically designed and developed the Evosep One for high-throughput applications, with a focus on clinical analysis of large sample cohorts. The newest and most powerful generation of mass spectrometers combined with novel acquisition strategies and software tools keep pushing boundaries and are now making comprehensive proteomes in short time a reality.
Here we evaluated the new ZenoTOF 7600 mass spectrometer with the novel ZenoSWATH acquisition strategy across the Evosep One standard methods. For high-throughput applications with 200 samples analyzed per day, we were able to reproducible identify and quantify more than 4,000 proteins from 200 ng HeLa digest. Collectively our results show that ZenoSWATH is a significant driver of deep proteomes, making the ZenoTOF 7600 an ideal match with high-throughput and standardized methods on the Evosep One.

Join this event to explore the power of single cell proteomics. The Evosep One is a powerful and standardized platform for high sensitivity work including single cell proteomics. The robustness and throughput of the Whisper methods on the Evosep One combined with the sensitivity of the Agilent 6495C triple quadrupole support targeted analysis of proteomes at the single cell level. We demonstrated this by targeting selected peptides in a complex background of sub nanogram material combined with dilution curves spanning several orders of magnitude. To ensure seamless integration of the EvosepOne with the Agilent 6495C triple quadrupole, we have developed a native driver for MassHunter and a robust solution for the Agilent nanoflow ion source.
