Whisper Zoom 40 SPD is a powerful method and recent developments exhibit a 20% improvement compared to the legacy method. ​
This is enabled by a refined flow strategy offering a more precise and confined sample delivery, which significantly improves the speed and overall sample recovery to collectively improve the sensitivity. ​
​Additional robustness compared to legacy methods is obtained from increased gradient flow at 200 nl/min, which gives you improved spray stability yielding more reproducible data. ​
​The required robustness is accelerated by the Evotip, a high efficiency and disposable trap column with no carryover, providing innovative sample introduction and sensitive long-term storage.​

FULLY AUTOMATED PAC DIGESTION GIVES COMPREHENSIVE PROTEOME COVERAGE WITH WHISPER ZOOM
Proteomics is undergoing a significant shift and rapidly evolving towards more efficient and user-friendly technologies, that can handle larger quantities of samples. This transition will require sustainable and cost-efficient strategies and as an example, a miniaturized workflow with less sample starting amount, will reduce enzyme usage and sample preparation cost. ​
This strategy will benefit from complete automation and direct integration with Evotip loading to utilize nearly 100% of the peptide digest and from the Whisper Zoom methods, which will allow you to reduce the sample input to just a few hundreds ng protein lysate, as exemplified by using the timsTOF HT mass spectrometer.​

COMMERCIALLY AVAILABLE SAMPLE PREPARATION WORKFLOW FOR SCALABLE SINGLE CELL PROTEOMICS
PROTEOCHIP: AUTOMATED SIMPLE, SEAMLESS SAMPLE PREPARATION
Sample collection, preparation, and transfer are often the primary sources of sample losses in high sensitivity proteomics applications.
To address this, a prototype chip, proteoCHIPâ„¢ Evo96, compatible with the Cellenion cellenONE instrument has been developed. The chip enables an automated workflow for direct transfer of peptides into Evotips and removes manual handling steps.
This workflow combines sample preparation with the Cellenion platform, pipetting-free transfer of single cell protein digests into Evotips, and analysis with the Whisper Zoom 40 SPD method together with the Bruker timsTOF SCP mass spectrometer.
High sensitivity is required in all steps of a workflow and this solution demonstrates excellent proteome coverage for single cells, with up to 4000 protein groups identified per single cell.
IT’S ALL IN THE EVOTIP
The Evotip, a disposable trap column, is the core of our technology. The Evotip serves as an integrated purification step, where contaminants are left on the tip, and analytically relevant peptides are eluted.
The Evotip has proven to be extraordinary important for ultra-sensitive analysis, as very small sample amounts can be loaded in higher volumes.
Furthermore, loaded Evotips can be stored cold until analysis, which is especially important for single cell proteomics, where very limited sample amount sets higher demands to the storage of peptide and become a crucial factor for reproducible analysis.
One tip – A simple, streamlined loss less one pot sample preparation workflow for proteomics
To accommodate the demands of single cell proteomics, the Olsen group have introduced the One-Tip protocol, a simplified workflow using Evotip Pure. The Evotip serves as a sample storage and introduction device for the Evosep One. By integrating cell lysis and digestion on the Evotip directly, you get a simple one-pot workflow for proteomics, as no further sample handling is needed.
Coupled with the Whisper Zoom 40 SPD method, they benchmarked the sensitivity and reproducibility of the One-Tip methodology by varying the number of HeLa cells from 20 to 3,000 cells.
The simplicity of the workflow makes it easy to automate on liquid handling robots and to become a new cornerstone for sensitive and high throughput proteomics.

AUTOMATED SINGLE-CELL SAMPLE PREPARATION FOR PROTEOMICS with Evotip Pure
In this publication, the Broad Institute presents a fully automated single-cell sample preparation workflow. The streamlined Evotip Pure protocol integrates the proteoCHIP EVO 96 with the Evosep One system, enabling in-line desalting and reproducible separation, doubling protein identifications without manual handling.
Combined with the Bruker timsTOF mass spectrometer, this method improves protein identifications two-fold over standard chromatography, identifying up to 4,000 proteins per HEK-293T cell. The protocol also profiles hundreds of LPS-perturbed THP-1 cells, identifying key regulatory proteins in interleukin and interferon signaling.
