High-sensitivity phosphoproteomics for high-throughput cell signaling studies

Protein phosphorylation status is important for understanding the cellular signaling state and key to exploit the potential of many kinase-targeting small molecule inhibitors. Alongside efforts to establish simple, integrated and automated workflows for robust and sensitive phosphoproproteomics profiling, shorter LC-gradients hold the potential to leverage scalable workflows. Here, we performed a phosphoproteomics evaluation of new and 2-3 times faster Whisper gradients and compared them to the Whisper 40SPD standard. Moreover, we applied the faster gradient to analyze phosphoproteome perturbation responses of a range of chemotherapeutic agents in triple-negative breast cancer cells.
