INTEGRATED PURIFICATION AND SEPARATION DELIVERED BY THE EVOSEP ONE
The Evosep One is a standardized platform designed for robustness and high throughput. This is achieved by using Evotips as disposable trap columns, where samples are loaded and desalted offline. The autosampler picks up the tip and integrates elution with liquid chromatography. This removes sample handling steps and reduces injection cycle overheads.
Minimal wear and tear on mechanical components is ensured as all elution and gradient formation happen at low pressure. The Evotip is left for disposal after each run, which then allows partial elution to extend column lifetime. Analytically important peptides are effectively retained, and cross-contamination reduced.
SINGLE CELL PROTEOMICS IS A ROBUST STANDARDIZED APPLICATION
Whisper100 40SPD beta is the fastest method in a series of methods developed with Whisper Flow Technology. It has a gradient flow of 100 nl/min with a 30 minutes method duration – delivered robustly by standard Evosep One hardware.
The method is ideal for ultra-sensitive applications, such as single cell proteomics, where one of the main challenges is to maintain sensitivity and robustness throughout the workflow.

This combination is extremely robust as only 20 bars increase in system backpressure was observed after thousands of runs as well as a reproducible flow profile.
IT’S ALL IN THE EVOTIP
The Evotip, a disposable trap column, is the core of our technology. The Evotip serves as an integrated purification step, where contaminants are left on the tip, and analytically relevant peptides are eluted.
This minimizes cross-contamination and extends column lifetime. The Evotip has proven to be extraordinary important for ultra-sensitive analysis, as very small sample amounts can be loaded in higher volumes.
Additionally, loaded Evotips can be stored cold prior to analysis.

One of the greatest goals of single-cell proteomics is to reach the needed sensitivity and maintain the performance throughout the lifetime of a project.
Andreas-David Brunner from the Mann group at the Max Planck Institute of Biochemistry has developed a robust workflow combining miniaturized sample preparation, the Whisper100 40SPD beta method and a novel trapped ion mobility mass spectrometer.
This workflow allowed the accurate quantification of proteomes and their changes up to a depth of 1,300 proteins when loading single, FACS-isolated cells.
