Global Analysis of Protein Expression and Turnover dynamics in Single Cells

Talk by Prof. Jesper V. Olsen, Novo Nordisk Foundation Center for Protein Research, University of Copenhagen

We introduce a streamlined single-cell proteomics (SCP) workflow for routine quantitation of >5000 proteins per cell. The workflow incorporates single cell dispensing and sample preparation using the cellenONE with a proteoCHIP EVO96 and direct transfer to Evotips. Subsequent LC-MS/MS analysis with Evosep One LC using Whisper flow gradients coupled to narrow-window data-independent acquisition (nDIA) on the Orbitrap Astral mass spectrometer provides throughput of up to 120 SPD samples per day using the new Whisper Zoom methods. Employing pulsed SILAC, we investigated global protein turnover in >1000 single cells from undirected differentiation of hiPSCs.

Accelerated Kidney Aging induced by Irradiation in Non-Human Primates analyzing Cortex and Medulla by Deep Proteomic Profiling

Chronic kidney diseases have emerged as problematic diseases during human aging. We investigated a model system in non-human primates (NHP) that were exposed to irradiation (IR) and that developed kidney disease and accelerated aging phenotypes. We used data-independent acquisitions (DIA) on a timsTOF HT with Evosep One HPLC and quantified >5,400 proteins (with 2 peptides each) to gain molecular insights into how IR exposure may drive renal aging processes in rhesus macaques.
