Plasma proteomics at scale with Evosep Eno – from discovery to target verification

Talk by Stoyan Stoychev, Senior Scientist at Evosep

The Evosep Eno is designed for robustness, standardization, and ease-of-use, offering significant enhancements in chromatographic performance. In this presentation, we highlight the advantages of incorporating the system into both shotgun and targeted proteomics workflows. When combined with fully automated sample preparation, the Evosep Eno enables deep proteome profiling at high throughput – up to 500 samples per day – without compromising quantitative precision or biological information. In targeted applications, the Evosep Eno demonstrates high sensitivity, detecting analytes at low amol levels with a linear dynamic range spanning 4 to 5 orders of magnitude. Furthermore, when paired with fully automated plasma sample processing, it enables accurate and precise, large-scale quantification of plasma targets.

Exceeding 100 spatially-resolved proteomes per day: An optimized ultrasensitive tissue proteomics workflow

Spatial proteomics (SP) enables precise mapping of protein abundance, localization, and interactions in tissues, offering deep insights into cellular function and disease. We co-developed Deep Visual Proteomics (DVP), integrating high-resolution microscopy, AI-driven image analysis, and laser microdissection with deep proteomic profiling. In this talk, I will present an optimized cellenONE protocol for loss-minimized tissue processing, benchmarked across all Evosep ONE Whisper Zoom gradients and three DIA methods on the timsUltra AIP. Our results demonstrate the feasibility of acquiring over 100 high-quality spatial proteomes per day—paving the way for large-scale, translational SP studies.
