Evotips

Evotip Pure is a consumable, C18 based device that serves multiple purposes. Peptides are immobilized onto the Evotip resin, desalted and stored until ready for measurement. Evosep Eno then elutes peptides from the Evotip Pure into the system as a start of separation process. Evotip is finally disposed of, together with unwanted contaminants. In short, Evotip Pure is a desalting device, a storage vessel for peptides and a single use, disposable Evosep Eno pre-column, all in one. Only Evotip Pure are compatible, tested and validated for use with the Evosep Eno. Evotip Pure are designed for a perfect low-pressure, leak free fit with the Evosep Eno. Use of any other product for sample introduction is not permitted and will void instrument warranty. Peptide solutions or other liquid proteomic samples are to be loaded on Evotip Pure according to the Evotip loading protocol. Find information here at www.evosep.com/evotip .

No, they are a single use consumable. Once the sample has been eluted from the Evotip, the plastic will be slightly deformed and cannot be used again. Doing so risks instrument damage and will void instrument warranty.

Evosep recommend following the Evotip loading protocol exactly and loading 20µl of sample. We have tested and were unable to detect losses from diluting a low volume sample to 20µL for loading on Evotip Pure. If the user chooses to change the volume the loading protocol would need to be adapted accordingly but this is not recommended.

The stationary phase of the Evotip Pure is an Evosep proprietary C18 resin.

Support and Maintenance

The User Manual contains details on the required maintenance. In short, the maintenance requirements are to empty, flush and refill solvent bottles with fresh solvents weekly. This should be followed by the Solvent exchange procedure. The Evosep Eno and Evosep One require a yearly Preventive maintenance visit, that are included in our service plans or can be purchased directly from Evosep. See our offerings here.

Evosep systems are built to be robust and easy to use. Most troubleshooting and maintenance can be performed by the user independently with our remote support without requiring an engineer visit. The Evosep interactive online troubleshooting guide  will help you identify the issue, and the Advanced User guide at www.evosep.com/support/documentation  and our video tutorials will show you how to do the repairs and replace parts when needed.

To ensure the fastest possible response from our team, the best way to reach us is to send an email to support@evosep.com. Priority is given to users with an Evosep service contract. Ideally, you would have already done all 5 diagnostic tests (as explained in Diagnostics section in Troubleshooting) and attached the Evosep logfiles (Advanced User Guide, chapter 12.1) in the email. There you can describe the issue and all the steps already taken. In many cases, we can help you solve the issue in the same day, and you can continue running your samples.

System and Methods

During installation users are trained on the Evosep Eno or Evosep One, and with this training users tell us they are able to start analysing samples even the same day.

Equivalent information is available also for the Evosep One.

Although the name and throughput of the Evosep Eno methods match those for the Evosep One, the methods with their set columns have been optimized for each instrument and are not identical. Evosep Eno offers improved chromatographic performance compared to Evosep One, see the brochure and method application notes for details https://www.evosep.com/evosep-eno/. Note that a column oven is required for all Evosep Eno methods and selected Evosep One methods.

Based on the type of proteomic analysis, timeframe and depth of coverage Evosep Eno has a range of methods of different throughput. You can select the method most appropriate for your analysis, but you cannot make adjustments to the set, standardized methods. The methods and their set columns are already optimized for peptide analysis to give a standardized result every day, every lab and every time.

Evosep has optimized their Evosep Eno methods for the specified columns for each method. This means that the best peptide chromatography performance and robustness will be achieved with the specified columns. Uniquely, Evosep guarantees the performance of the system with the specified column matches their method application note for a digest standard and supports users to achieve this. Evosep cannot guarantee performance or support any other columns. Both Evosep One and Evosep Eno methods are optimized using LC/MS grade water with 0.1% formic acid as solvent A and LC/MS grade Acetonitrile with 0.1% formic acid as Solvent B. Use of any other solvents are not supported and may damage the system.

No, direct infusion of a sample is not possible with the Evosep Eno or Evosep One instrument.

Yes, both Evosep Eno and Evosep One can easily be connected to different types of MS systems from multiple vendors. You can find more details at www.evosep.com/support/evosep-one-compatibility and you can download necessary software from www.evosep.com/support/software-updates. 

Evosep guarantee and support users to get chromatographic performance matching Evosep’s method application notes for a standard protein digest sample. If desired this performance can be used for performing regular QC of the system. For formal qualification of the Evosep One an operational qualification (OQ) is recommended annually. Speak to your local representative for more details.

Peptide solutions or other liquid proteomic samples are to be loaded on Evotip Pure according to the Evotip loading protocol on which they are desalted and stored until ready for analysis. The loaded Evotips are eluted by the system. Only Evotip Pure are to be used with the Evosep Eno or Evosep One, use of any other product for sample introduction will void instrument warranty.
